Immunoprecipitation and mass spectrometry identify non-cell autonomous Otx2 homeoprotein in the granular and supragranular layers of mouse visual cortex

Neuronal circuits generated during embryonic development are remodelled during critical periods in postnatal development. Otx2 homeoprotein helps determine the timing of these critical periods by signalling fast-spiking parvalbumin (FSPV) interneurons in mice. Otx2 in the visual cortex is non-cell autonomous and is taken up by FSPV cells. Because Otx2 and Otx1 may have redundant functions, Kim et al. set out to investigate whether Otx1 also plays a role in the timing of critical periods and whether part of the Otx staining observed could be attributed by Otx1 rather than Otx2. They found that while Otx1 is expressed in the cerebral cortex, the Otx staining is maintained in Otx1-null mice and that immunoprecipitation in layers of the visual cortex showed bands corresponding to Otx2 and not Otx1.
