In this study Fameli and colleagues demonstrate that nanojunctions between lysosomes and the sarcoplasmic reticulum couple cellular Ca2+ waves to lysosomal activation. Using tomographic transmission electron microscopy to characterize these junctions, and simulations of NAADP-dependent junctional Ca2+ transients, they demonstrate that the number of observed junctions is sufficient to generate a Ca2+ signal above the threshold required for Ca2+-induced Ca2+ release and that these junctions are a pre-requisite for efficient Ca2+ signal coupling.
