In this study Fameli and colleagues demonstrate that nanojunctions between lysosomes and the sarcoplasmic reticulum couple cellular Ca²⁺ waves to lysosomal activation. Using tomographic transmission electron microscopy to characterize these junctions, and simulations of NAADP-dependent junctional Ca²⁺ transients, they demonstrate that the number of observed junctions is sufficient to generate a Ca²⁺ signal above the threshold required for Ca²⁺-induced Ca²⁺ release and that these junctions are a pre-requisite for efficient Ca²⁺ signal coupling.
